Alizarin Red S. technique and its application in veterinary sciences: A review
Abstract
Alizarin Red S (ARS) is a primary calcium-binding dye utilized in veterinary sciences to visualize mineralized bone and calcium deposition in whole-mount skeletal preparations, histological tissues, and osteogenic cell cultures. This review outlines the fundamental chemical principles, laboratory workflow, and technical protocols associated with ARS staining, including specimen preparation, staining, washing, clearing, and mounting. By highlighting primary diaphysis and secondary epiphysis ossification sites across various developmental stages, ARS serves as an essential bridge connecting veterinary anatomy, embryology, radiology, and orthopedics. Understanding these normal skeletal mineralization patterns enables clinicians and researchers to accurately distinguish normal developmental structures from fractures, physeal injuries, or congenital skeletal anomalies in immature animals. Furthermore, the review addresses key visual observations, methodological limitations, common technical artifacts, and strategies for protocol optimization in developmental research and surgical education.
