Black Pepper (Piper nigrum): The King of Spices: A Comprehensive Review of Its Phytochemistry, Pharmacological Activities, and Therapeutic Potential

Abstract

Background: Black pepper (Piper nigrum L.), belonging to the family Piperaceae, is one of the most widely used spices worldwide and is often referred to as the “King of Spices.” It consists of the dried, unripe fruits of a perennial climbing plant cultivated mainly in tropical regions such as southern India, Indonesia, Brazil, the Malay Archipelago, and the West Indies. Black pepper holds an important place not only as a culinary spice but also in traditional systems of medicine such as Unani and Ayurveda. It is known by various vernacular names, including Kali Mirch (Hindi/Urdu), Pippali (Sanskrit), and Milagu (Tamil). Objective: This review aims to provide comprehensive and updated information regarding the botanical description, phytochemical constituents, pharmacological activities, and therapeutic uses of Piper nigrum, particularly in Unani medicine. Methods: A review of available literature was conducted using various scientific sources, including research articles, books, and traditional medical texts related to the botanical, phytochemical, and pharmacological aspects of Piper nigrum. Results: The major active constituent of black pepper is piperine (1-piperoyl piperidine), which is responsible for its pungent taste and many of its biological activities. Piperine exhibits a wide range of pharmacological properties such as antioxidant, anti-inflammatory, antimicrobial, antitumor, antiasthmatic, antipyretic, analgesic, hepato-protective, Immunomodulatory, anti-depressant, and digestive-stimulating effects. It also enhances pancreatic and intestinal enzyme activity, thereby improving digestion and nutrient absorption. The fruits of Piper nigrum are processed to produce black, white, and green pepper and are widely used as flavoring agents, preservatives, and in traditional medicinal preparations. Conclusion: Piper nigrum is a valuable medicinal plant with significant therapeutic potential. Its diverse pharmacological activities, mainly attributed to piperine and other bioactive compounds, support its traditional use in Unani medicine and highlight its importance in modern pharmacological research.
