Utilization of microbial lipases for bone degreasing in Gelatin preparation

Abstract

One of the main basic materials used to prepare gelatin is the cattle bone. There are numerous additional biological contaminants, such as leftover protein and fat, in the crushed bones. In order to determine the quality and characteristics of gelatin that would satisfy the intended purpose, collagen purity is crucial. Purified collagen is typically obtained by removing unwanted contaminants through chemical and physical pre-treatment. Regular chemical pre-treatment has the drawbacks of causing environmental contamination and bone deterioration. The employment of enzymes lessens the adverse effects on the environment by lowering the amount of chemicals, energy, wash water and waste that is subsequently produced. These days, a wide range of industries use lipases extensively as degreasing agents. There were three stages of the study viz., Temperature optimisation for incubation; concentration of lipase; microbial lipase incubation period, leading to the development of a stan for the lipase enzyme-based degreasing of crushed bone. The effectiveness of fat reduction percentage before and after enzyme therapy was essentially the basis for selecting the ideal conditions throughout the investigation. The results indicated that incubation temperature of 47.5?C, lipase concentration of 1% of bone weight and incubation duration of two hours had significantly beneficial effect on lipase activities in degreasing of crushed bone. Using the standardised procedure, the fat content of bones was found to be reduced by 22.42% when subjected to the action of microbial lipase. Thus, the enzymes lipase can be used as one of best option for degreasing of the crushed bones.
